For a misfire limited to a particular cylinder (as opposed to random misfire between all cylinders): 1. Bad/worn/contaminated spark plug 2. Ignition coil failure (coil-on plug) (often a concequence of running the engine with worn spark plugs) Those two are by far the most common reasons. Much more rare: 3. Injector failure 4. Leaking/burnt valves 5. Failed valvetrain components (lifters, camshaft lobes, pushrods, valve springs, rocker arms) 6. Leaking head gaskets (actually, resulting in spark plug fouling by calcium deposits) 7. Damaged pistons, conrods or bearings 8. Manifold vacuum leaks (injector seals, although sometimes manifold gaskets too) 9. Wiring problems for injector or coil circuit (including internal PCM/control module issues) In about that order, resulted on 30 years of experience with different makes and models All other stuff mentioned here is rather a fuel mixture/fuel trim issues that might be resulting in random misfire as a consequence of too lean or too rich mixture.

People come in frequently during weather shifts, thinking they need a patch or new tire because their tpms light is on. #1 the valve core inside the valve stem (the thing that you put the air hose on to fill tires) can back itself out with temp changes causing a slow leak, so get a valve core tool from autozone for your glove box to tighten them if needed. #2 usually its not just one low tire, they're all low from temp changes & customer's happy they didn't need a tire. #3 tpms sensors have little batteries in them, which one dying can be sped up by temperature shifts. So when the tire shop tells you your sensor is dying & the tires aren't actually low, you need a new sensor installed in the wheel, as the batteries cannot be replaced. Usually when it's just a bad sensor & not actual low tire pressure, the tire light will flash a couple times when you first turn the key. Thats the systems way of alerting you to the bad sensor. If it doesn't flash at all, it may be a pressure issue, too high or too low.

I always tell my wife whenever she gets a patched tire donโt ever go over 85 mph. The tire is already partially damaged and repaired and can rip the patch off over any extended distance. The speed ratings basically only apply to a tire in good condition.
