Hiroshima was chosen because it was a "virgin target" IE it hadn't been bombed, being not militarily important. And also it has a very distinctive Y-shaped river leading into harbor formation, that could be made out even under some cloud cover. It was known as a city of temples, and also as a city where a lot of refugees of other bombed-out cities had gone to. Other than the small-time cottage industry that was everywhere in Japan, the city didn't have much to do with the war effort.
