Colonies - as a historic concept - were never inhabited places, but were places were already other people live. When we crossed the Bering Strait, we did not colonize that territorory (America) but humanity expanded there. Also, humanity never expanded or settled on Antarctica due to unlivable conditions, we only have some outposts there, similar in nature as a space station. So there is no reason to assume we will ever expand to Mars to really settle there, we could only achieve setting on Mars as an outpost, partly but not completely independend from earth. If we ever want to transform Mars into a place were humans can live (but there is no real reason for doing that) you can't reach that by sending humans - there usable worktime will be occupied for 99% by the requirements of staying alive, water, oxygen, food, settlements, protecion agains radio-active rays and dust) - we could better develop self supporting robots with artificial intelligence to exploit the natural resources of Mars, to reproduce themselves and evolve over time, which can also manufacture the resources (water, energy, oxygen, shelter, air-pressure, protection against cosmic rays and dust, and food) so humans can go there and do other tasks.
