  2. You can extract DNA/RNA from a cut-out section of an agarose gel by dissolving it.

  3. 3:05 & 3:15. The left hand lane contains DNA of fixed sizes to use as a scale. In checking correct insertion you can run a lane with the unaltered plasmid as a comparison or (better) digest some of the new plasmid and check to see if the right size insert appears.
